Preliminary Testing at TRIO
Welcome to TRIO Fertility, and thank you for taking the first step in your fertility journey with us. After your initial appointment, your physician has recommended specific tests to help us build a clear and personalized plan for you. Below, you’ll find everything you need to know about these tests, including instructions, timing, and next steps.
As your doctor mentioned, testing is based around the timing of your cycle. Day 1 is the first day of your period, full flow – not spotting.
Please email both the Day 1 team (day1@triofertility.com), along with cc’ing your doctor’s office on the first day of your next period, and we will help to arrange the following tests at TRIO:

Frequently Asked Questions
What is the purpose of fertility testing?
Fertility testing helps your TRIO care team understand your baseline fertility profile so your physician can recommend the most appropriate next steps and create a tailored plan. Learn more:Â https://triofertility.com/intro-fertility/
What tests might be included in my initial workup?
Common testing may include bloodwork (hormone testing), ultrasound (to assess ovaries and follicles), and—when applicable—sperm testing via semen analysis. Your physician will confirm which tests apply to you.
When should I do my bloodwork?
Some blood tests are typically completed early in the menstrual cycle (between days 2-5) to assess hormone levels that relate to egg supply and ovulation. Other testing can be done at any time. Follow the timing provided by your care team.
Do I need to fast for my bloodwork?
In some cases, fasting bloodwork may be required. If fasting is required your doctor will let you know.Â
Why do I need a urine sample for my testing appointment?
Some new patient testing requires a urine sample. You’ll be asked to arrive early to collect a specimen jar from the blood lab before proceeding to your ultrasound appointment.
What should I expect at my ultrasound appointment?
Your ultrasound will include two steps:
- a full bladder transabdominal ultrasound, followed by
- a transvaginal ultrasound after you empty your bladder.
Do I need to bring anything with me to my appointment?
If you are doing your testing at a TRIO location we will have your bloodwork requisitions on file. If you are doing bloodwork at an outside lab you will need to bring all requisitions with you to ensure all tests are completed. You also may be asked for a copy of your ultrasound requisition, which your medical team will email to you.
What is a sonohysterogram, and how do I prepare?
A sonohysterogram is a diagnostic procedure used to evaluate the uterine cavity and may also help assess whether the fallopian tubes are open. If scheduled, you may be advised to take pain relief (e.g., Advil or Tylenol) about one hour before. You will also be given antibiotic with instructions to start it before the procedure and complete the prescribed course.
Are there any restrictions before a sonohysterogram?
You must abstain from unprotected intercourse after your cycle begins until the sonohysterogram is completed.
What is a semen analysis or DNA Fragmentation testing (for patients with sperm), and why is it done?
A semen analysis is commonly used to evaluate sperm number, movement, and shape to help rule out sperm-related factors that may affect conception.A DNA fragmentation test is a specialized laboratory analysis that measures the percentage of sperm cells with damaged or broken DNA strands, evaluating genetic quality beyond a standard semen analysis. Both tests can be done to identify causes of unexplained infertility, recurrent pregnancy loss, and failed IVF/ICSI cycles, helping clinicians choose the best treatment path.
Who should I contact if I have questions while I’m completing testing?
During the testing phase, please direct questions to your physician’s administrative team (contact details listed on this page). This helps ensure the right team can coordinate your testing, answer process questions, and support next steps efficiently.
